Milenia HybriDetect New Citation Record! Citation List Update

HybriDetect joined the world of science in 2006 with the paper “DNA Detection Using Recombination Proteins” by Piepenburg et al., in which they presented their invention of Recombinase Polymerase Amplification (RPA). Since then, much has happened in the world of lateral flow and nucleic acid analysis.

The Milenia HybriDetect product line (HybriDetect und Milenia HybriDetect 2T) has been successfully combined with a variety of methods, such as PCR, RPA or LAMP. The discovery of the CRISPR/Cas system (Clustered Regularly Interspaced Short Palindromic Repeats/CRISPR-associated protein) by Charpantier and Dougna also influenced the use of HybriDetect a lot. But before we jump to the application methods we have one question to discuss:

How Does The HybriDetect Testline Perform Over The Years?

As you can see in Figure 1, after initial low publication numbers due to low awareness, the HybriDetect product line has been well established as an evaluation platform which is widely used by scientists and companies all over the world.

Today Milenia HybriDetect is frequently cited in more than 285 peer reviewed publications, including high impact journals such as Nature or Science.

The excellent suitability of HybriDetect for detecting viruses such as Ebola, Zika and SARS-CoV-2 has further accelerated this positive development in recent years. We are pleased to announce that once again we will be able to record a new maximum number of annual publications already in early November.

Application Fields Of HybriDetect

This year the focus is not only on the Covid-19 pandemic as in 2020/21, but also on the detection of other pathogens, increasingly in the veterinary and agricultural sectors (Figure 2). The HybriDetect can be used wherever rapid diagnostics at point-of-care is needed. In the field of public health it can be used not only for the detection of pathogens, but also for the detection of cancer cells, mutations and SNPs (Single-Nucleotide Polymorphism). The identification of the sex of animals and plants in the veterinary and agricultural sectors was also shown this year. These are only a few examples of the areas of application. More detailed information can be found in the blog post Milenia HybriDetect – Type of Use & Fields of Application.

Molecular Amplification Methods And HybriDetect

The application of HybriDetect is versatile. Figure 3 gives a brief overview of the methods used in conjunction with the HybriDetect in 2022. Mostly isothermal methods were used to enable application at point-of-care.

More than 90 % of all papers citing Milenia HybriDetect use isothermal DNA amplification.

This year, research was focused especially on combining the HybriDetect platform with RPA (40%) or CRISPR/Cas (44%). Other methods were also used in 2022 citing HybriDetect, of which the RAA (recombinase aided amplification), RCA (rolling circle amplification) and the use of aptamers look promising.

Further information on how the individual methods work in combination with the HybriDetect Universal Test Strip can be found here:

CRISPR/Cas – A Combination Of Methods

Frequently, the CRISPR/Cas-HybriDetect method is combined with a preceding RPA or LAMP (Figure 4). The aim is to amplify the gene sequence in order to increase sensitivity. CRISPR/Cas then serves to modify the amplified product so that it can be evaluated on the HybriDetect. In very few cases, CRISPR/Cas is used as a stand-alone method (14%).
