myilab research – Lateral Flow Reader

Since 2020, lateral flow rapid tests have been accepted and used by the wide public as a tool for diagnosing diseases and pathogens. Lateral flow tests can provide answers to questions in a wide variety of areas. Is the patient infected with HBV (Hepatitis B Virus)? Is the pig herd free of ASF (African Swine Fever)? Is the cultivated grain infected with wheat yellow rust or not? For all these tests a yes/no-answer is generated.

Quantification of these results is often not possible but can be interesting for
many applications and especially in the development of assays.
Milenia Biotec has addressed this issue and is pleased to announce the release of
the Lateral Flow Reader myilab research for the use in our Lateral Development
Platform HybriDetect.

 

For years, Milenia Biotec has enabled scientists and companies to develop their own detections with the universally usable evaluation platform HybriDetect, which includes the following products:

 

These are rapid tests that generate a yes/no-answer, but whose signal strength also correlates with the amount of analyte. Therefore, a semi-quantitative evaluation is possible by using evaluation cards such as the one shown in Figure 1.

Although this method allows a rough type of quantification, the visual evaluation of the test result is difficult, especially in borderline areas. Therefore, the result can depend on the evaluator and may be assessed differently by different people.

A frequently used alternative for quantification is the readout of images with image processing programs like “ImageJ”. However, this multi-step process is time-consuming, imprecise, and it can be difficult to compare multiple images under different lighting conditions. Many customers and developers have asked for a simple and reliable method of quantification. To meet this need for objective and reproducible quantification, Milenia Biotec has developed its own readout device, the myilab research (Figure 2).

Compatibility

The myilab research is a universal dipstick reader that can be used to quantify all our HybriDetect products. The HybriDetect Cassette (MGHC 1) or the myilbab research adapter can be inserted directly into the reader (Figure 2A). The HybriDetect (MGHD 1) and HybriDetect 2T (MGHD2 1) test strips can also be measured directly without any intermediate steps thanks to the adapter (Figure 2B).

Time and Handling

It takes 10 seconds from the initial start-up of the device to the first measurement. The measurement time of a single test is 15 seconds plus a few seconds for the insertion of the test cassette or the insertion of the test strip into the adapter. All combined less than 30 seconds are required for a complete measurement cycle from insertion of the test strip/cassette to result display. The device is intuitive and easy to use.

Evaluation

The results of the measurement are shown immediately after the measurement on the display for each line. The position of the detected lines is also shown on an enlargeable image section. This allows you to control your result. The results can be exported via e-mail as a csv-file for further processing.

The first slide shows the result of the first line. The last slide shows the intensity of the control line.

Example Scenario

The following diagram shows the correlation between the concentration of the analyte (in this case a biotin/FAM-labelled ssDNA oligonucleotide) and the signal intensity of our HybriDetect cassette measured with the myilab research indication.

myilab research vs. Visual Evaluation

The human eye is ideally suited to give a yes/no-answer to a signal. Therefore, it is interesting to see how the myilab research compares to the evaluation by eye regarding a yes/no-answer. The following table shows this comparison for the products MGHD 1, MGHD2 1 and MGHC 1.

Except for a limit concentration of 0,001 pmol/LFA, all analyte quantities that are assessed as positive by eye can also be reliably detected as positive with the myilab research. Optically negative results are also displayed as negative by the device. In addition, unlike the human eye, the myilab research provides reliable quantification as soon as a signal is detected, allowing conclusions to be drawn about the analyte concentration.
